采用 Gateway® 技术的 ProQuest™ 双杂交系统 (ProQuest™ Two-Hybrid System with Gateway® Technology) 是一种基于酵母的体内系统,用于鉴定两种蛋白之间的相互作用。 这种相互作用会重构功能性转录因子,该转录因子激活整合在染色体上的报告基因,而转录激活与否可通过选择性培养基上的细胞生长情况来进行监测。

ProQuest™ 系统 (ProQuest™ System) 具有如下特点:

a)      采用 Gateway® 技术,能够快速简便地生成“诱饵”和“猎物”构建体,有利于下游应用

b)      低拷贝 DBD(DNA 结合结构域)和 AD(活化结构域)载体 (ARS/CEN),可控制过量表达,提高重复性

c)      三种不同报告基因(HIS3、URA3 和 lacZ),具有独立的启动子区域,可快速筛除假阳性。 URA3 报告基因可用于阳性选择和阴性选择,从而可以使用高级双杂交技术,例如反向双杂交。

 

d)         一组由强到弱、基于 Gateway® 的相互作用对照组,用于评估实验结果以及实验组相互作用的强度。
