pUC118载体简介:

pUC118载体是利用pUC18载体的Nde I 酶切位点,插入含有M13 phage DNA Intergenic region (IG) HgiA I (5645) - DraI (5941) 片段构建而成的。由于在lacZ基因中含有多克隆位点,因此在含有IPTGX-Gal 的平板培养基上可以很容易地通过蓝白筛选判断载体中有无DNA片段的插入。同时还可以通过载体上的lac promoter表达外源基因,使用M13系列的通用引物对插入载体中的DNA片段进行测序。使用Deletion Kit for Kilo-Sequence (Code No. : 6030) 可以对克隆得到的数千碱基的DNA片段进行测序。
由于pUC118载体中插入了M13 phage DNAIntergenic region (IG),因此pUC118载体还可以在Helper phage M13K07的感染下产生单链DNA,包入Phage颗粒内后从菌体内放出,而不存在Helper phage单链DNA的污染。使用本制品时,可以稳定获得单链的长片段DNA (~7kb),并且没有缺失。
